If I'm enrolled in autopay, and I have enough money on my account when the bill is due to cover the plan amount, will my credit card be billed?

No.  Money should be taken from your account balance first.

Your card is charged only to refill your balance 1 to 2 days before your plan renews.  On the renewal date that money is used and your balance is zero again.
